چکیده

A microscopic mass balance approach has shown that the initial saturation (Is), absorption number (An), dose number (Do), and dissolution number (Dn) are four fundamental dimensionless parameters that can be used to estimate the fraction dose absorbed (~ of suspensions of poorly soluble drugs in humans. The dissolution number of a drug increases with decreasing its particle size. The effect of micronization on F for suspensions was investigated in terms of Dn. About 90% of maximal Fcan be achieved at Dn-~2. Increasing the solubility of a drug results in better oral absorption through increasing Dn and decreasing Do. The fractions dose absorbed of digoxin, griseofulvin, and benoxaprofen agree with predicted F values using estimated parameters. Drugs with low Do and low Dn can be more completely absorbed by reducing particle size, while absorption of drugs with high Do and low Dn is limited by solubility and requires higher solubility to enhance the fraction dose absorbed in addition to micronization. Solubility at the physiological pH should be used for the estimation of the fraction dose absorbed.

Meloxicam transdermal delivery: effect of eutectic point on the rate and extent of skin permeation

Objective(s):Drug delivery through the skin can transfer therapeutic levels of drugs for pharmacological effects. Analgesics such as NSAIDs have gastrointestinal side effects and topical dosage forms of these drugs are mainly preferred, especially for local pains. Meloxicam is one of NSAIDs with no topical form in the market. In this research, we attempted to quantify the skin permeation of a m...

THE EFFECT OF ADRIAMYCIN ON SALT FRACTIONATED CHROMATIN

In this study calf thymus chromatin was fractionated into active (S1 and S2) and inactive (P 2) chromatin. Then the interaction of an anthracyc1ine antibiotic, adriamycin, with these fractions was investigated employing absorption and difference UV Nis spectroscopy and SDS and agarose gel electrophoreses. The results suggest that the binding of adriamycin to the S 1 fraction is slight but ...

Effects of Nanoclay on Cellular Morphology and Water Absorption Capacity of Poly(vinyl alcohol) Foam

The present work was aimed to examine the effects of incorporation of each of two different types of nanoclay, i.e. Cloisite Na+ and Cloisite 30B, into PVA foam on cellular morphology and water absorption capacity. Foam samples containing 0.0-10.0 wt% of each of the two types of nanoclay alone were prepared using mechanical foaming. Accordingly, PVA/organoclay/water suspensions were prepare...

Micronization: a method of improving the bioavailability of poorly soluble drugs.

For poorly soluble drugs, the digestive absorption depends on their rate of dissolution. Decreasing the particle size of these drugs improves their rate of dissolution. Fine grinding mills are use to micronize powders: either jar mills or fluid energy mills. Theses processes were applied to griseofulvin, progesterone, spironolactone and diosmin. For each drug, micronization improved their diges...
